I was able to identify my bacteria as Pseudomonas Aeruginosa. I spent all of Monday morning just extracting DNA samples from my bacteria. The first extraction method I use was the TE Freeze and Thaw method. It is a simple procedure. A person must take 1mL of his or her bacteria and centrifuge it for a certain amount of time. Later boil the bacteria, then place it in ice water, boil again, and repeat the freezing and thawing cycle a few times, The goal is to broken down the components of the bacteria so it can lose DNA particles. Some tests are more effective than others, of course. Well, I got to keep this up because all those DNA are not going to extract themselves. Ciao.
Hi Barierane, were you able to successfully extract DNA using the methods you tested?
ReplyDeleteYes! The only issue is that some of the tests such as the Boiling method produce a low quantity and quality of DNA so IO wasn't so sure if the test was effective.
