The first official week of internship just started. I was given an unknown bacteria and was instructed to to do a Gram test and incubate it. The Gram test was far more difficult than I would like it to be. I must have done it at least four different times to get even a decent sample. I figured that I am not letting the chemicals sit on the surface of the glass slide long enough. When my bacteria finish incubating then I will try to get a more concentrated specimen to work with for the Gram test. Practice makes perfect. I was also given a lab notebook to document all my findings and everything seem pretty cool so far. I have high hopes for this project and hopefully I figure out what my bacteria is soon.

ReplyDeleteThe gram staining was difficult for me as well. I had a hard determining if the bacteria was gram negative or positive. However, I learned that if you are not sure that you can inoculate the bacteria in question on a MacConkey agar plate. If nothing grows, then it is gram positive.
