Blog Post Week 5



Today is the day I finally completed DNA extraction all Pseudomonas Aeruginosa using six different procedures. It actually took me some time to get the hang of doing all the different procedures, but it was pretty exciting nonetheless. It is nice knowing the fact that all the hours you spent on your project finally pays off when you get to see just how much DNA was yield. My experiment shown that the Lysis experiment supplied virtually no nucleic acid while the Meat Tenderizing produce the most nucleic acid. My hypothesis was wrong and I am not sure why I receive these results. Further researching will probably supply me the answers.

  1. What did you use for the lysis experiment? How did you measure whether or not you extracted DNA? Sometimes we cannot visualize a DNA pellet directly and have to analyze it using other methods. Good luck!

    1. I use a lysis buffer for the lysis experiment and follow similar steps for the experiment like that of the TE Boiling. I centrifuge for a while, add the lysis buffer, boil it, and centrifuge again mostly. To extract the DNA, I was fortunately enough to have very visual pellets. IF there is no pellet, I try to look for floating particles that are possibly DNA particles.

  2. Great! I am glad to hear you saw some pellets. Just keep in mind, no visual pellet does not mean there's no DNA ;)
